Analytical techniques for cell fractions. I. Simplified gradient elution programming.

Many methods have been devised for producing gradients in elution fluids for ion-exchange chromatography (l-9). While many of these are invaluable for special purposes, none appears to allow: (a) the production of a complex multicomponent gradient directly from a graphic presentation, and (b) the possibility of changing from gradient to stepwise elution with the same system. A simple method for...

High-performance liquid chromatography for ultraviolet-absorbing constituents in human urine.

Several liquid-chromatographic systems involving anion-exchange resins and linear acetate gradient elution have been reported previously for the analysis of ultraviolet-absorbing constituents in human urine. We describe a high-performance liquid-chromatographic system on which a macroreticular anion-exchange resin, and stepwise elution are used. Separations were more rapid with ammonium chlorid...

Silver ion chromatography using solid-phase extraction columns packed with a bonded-sulfonic acid phase.

Commercial solid-phase extraction columns packed with a stationary phase with bonded benzenesulfonic acid groups are readily converted to the silver ion form and can then be used for silver ion chromatography of lipids. To illustrate the utility of such procedures, methyl ester derivatives of fatty acids with zero to six double bonds were separated from each other by a simple stepwise elution s...

Modeling of Gradient Elution in Multicomponent Nonlinear Chromatography

-Gradient elution in nonlinear chromatography is a very powerful tool for the separation of components having a wide range of retentivity. It is not only used for chemical analysis, but also used in preparativeand large-scale chromatography for the separation of many macromolecules, such as proteins. In this work, a general rate model has been presented for the study of gradient elution in nonl...

Separation and quantitation of free fatty acids and fatty acid methyl esters by reverse phase high pressure liquid chromatography.

Reverse phase high pressure liquid chromatography (HPLC) on octadecylsilyl columns separates mixtures of either free fatty acids or fatty acid methyl esters prepared from mammalian tissue phospholipids. Acetonitrile-water mixtures are used for the elution of esters. Aqueous phosphoric acid is substituted for water for the separation of the free acids. Unsaturated compounds are detected and quan...
